Tiny Molecule Helps Control Blood-Vessel Development, UT Southwestern Researchers Find

The developing and touch on of heart tissue and blood vessels is intimately tied to a tiny piece of ribonucleic acid (RNA) that is launch nowhere else in the body, researchers at UT Southwestern Medical Center have found.

Because of its specificity to the cardiovascular system, this "microRNA" is an attractive potentiality target for therapeutic treatment, the researchers said.

"Manipulating this microRNA provides a totally new way of addressing cardiovascular disorders," said Dr. Eric Olson, chairman of molecular biology and senior author of a work appearing in today's issue of

MicroRNAs are midget snippets of genetic material, naturally produced by the body, that help tweak the production of proteins by DNA. More than 500 have already been identified.

In the current work, the researchers focused on a specific microRNA called miR-126, which was already known to be associated with blood vessels. They found that miR-126 is found exclusively in a class of cells called endothelial cells, which line the inside surfaces of blood vessels.

Endothelial cells control the development of new blood vessels in developing embryos; the repair of injured origin vessels; and the creation of blood vessels to support development tumors.

The researchers genetically engineered mice to lack miR-126, and found that about 40 percent of them died before or just after parentage. These mice showed cardiovascular abnormalities such as fragile, leaking lineage vessels.

The surviving mice, however, appeared normal and lived to adulthood. The researchers complete that miR-126 is important in creating new vessels, but one time the cardiovascular system is established, it is not needed to maintain the system.

However, the surviving mice were less able to recover from a simulated heart attack. Almost all mice wanting miR-126 died within threesome weeks, while 70 percentage of normal mice survived for at least three weeks.

The researchers also tried and true the role of miR-126 in the branching of blood vessels using cut sections of mouse aortas in acculturation. When civilized with outgrowth factors that stimulate ramous, aortal sections from normal mice displayed branching of their endothelial cells.

Aortal sections from mice lacking miR-126, however, showed much less branching.

"MicroRNA research represents a new frontier in sympathy and treating human disease. This is just a hint of what can come," aforementioned Dr. Olson, director of the Nancy B. and Jake L. Hamon Center for Basic Research in Cancer and the Nearburg Family Center for Basic Research in Pediatric Oncology.

The researchers have filed various patents related to miR-126, and design to licence it for development as a curative agent through Miragen Therapeutics, a Boulder-based company co-founded by Dr. Olson, which UT Southwestern owns fairness in.

Other UT Southwestern researchers involved in the study were lead writer Dr. Shusheng Wang, post doc researcher in molecular biology; Dr. Arin Aurora, post doc researcher in molecular biology; graduate educatee Brett Johnson; Xiaoxia Qi, research scientist in molecular biology; John McAnally, research associate in molecular biology; Dr. Joseph Hill, prof of interior medicine; Dr. James Richardson, professor of pathology; and Dr. Rhonda Bassel-Duby, prof of molecular biology.

The work was supported by the National Institutes of Health, the Donald W. Reynolds Clinical Cardiovascular Research Center, the Sandler Foundation for Asthma Research, the Welch Foundation and the American Heart Association.

Jack Black leaves 'Man-Witch'

Would have been first project with 'Old School's' Phillips

Jack Black has fallen out of "Man-Witch," Todd Phillips' supernatural comedy that's set up at Warner Brothers.

When the star was attached last year, it was hailed as notable because it was the first collaboration between the boxoffice-drawing comic actor and the force behind such comedy powerhouses as "Old School" (which Phillips wrote, directed and produced) and "Borat" (which he wrote.)

"Man-Witch" centered on a man who discovers he's a witch and then heads off to teach at a female witch school; Phillips was set to produce and, likely, to direct.

Sources said that the decision this week to part ways may have originated from Black's camp, which had questions about Phillips' commitment to direct the picture ahead of "Hangover," another comedy said to be close to the helmer's heart. That project is also set up at Warners.

Warners confirmed the departure but declined to provide further detail. A spokesperson for Black said the star had no comment. A spokesperson for CAA, which reps Phillips, had yet to comment by press time.

The actor's departure has not affected the status of the movie, which remains in active development. Warners and Phillips are said to be combing a list of stars to replace the funnyman.

Black stars in the upcoming comedy ensemble "Tropic Thunder" as well as in the Judd Apatow-produced "Year One."

The Black-voiced "Kung-Fu Panda" was the top-grossing movie at the boxoffice last weekend, surpassing expectations by earning more than $60 million domestically.

T.I. Previews His Forthcoming LP, Paper Trail, For MTV News

The lyrics made it sound like T.I.'s pen was actually a match.

The King of the South was igniting each track with a viciousness that could well have fans and critics calling his forthcoming Paper Trail LP (due August 12) his best since Urban Legend.

Clearly, Tip's half-year-plus under house arrest wasn't just spent building a huge replica of the Eiffel Tower out of Legos ("It took me two straight days," he said of his work). The MC — who recently said he's not angry that he only came in at #10 on MTV News' "Hottest MCs in the Game" — found escapism in his music and, as he played a handful of tracks from his forthcoming album, at times he sounded better than we've ever heard him.

There are boasts such as "Without the braids, I'm the closest thing to O-Dog" and admonishment of peers like, "What the game need with you? They got me."

Yes, Tip is "back on top like a toupee."

"Turn My Beat Down" feels like a 2008 version of LL Cool J's "Boomin' System." The bass is turned all the way up and the synths inject a futuristic feel to it. Meanwhile, Tip experiments with his voice, but his message is clear: His sounds shake up the city.

"People see me in the street now/ And they wave and tell me to turn my beat down," he raps on the chorus.

"You're a-- in trouble if you pull up next to me/ Beat so loud, you can't hear yourself think," he adds in a verse.

"On Top of the World" is one of the strongest contenders for the album's first single. It stars the newest member of the Grand Hustle fam, B.O.B., on the hook and Kanye West on a guest verse.

"It's just a track of reflection," Tip said of the song. "Looking back to my humble beginnings, just reveling on how far I've come. It's definitely a record that people like Kanye and I ... and even B.O.B. [can relate to]: when you're on the outside looking in and you know you're good. You know you're worthy of being in the game. You know you can compete on a professional level. And once you prove that, it's a level of self-assurance. I can't even describe that."

Tip dances with self-doubt on "My Life, Your Entertainment."

"Kids, you don't wanna be like me/ It's safer to watch me on TV," he says in one line. Later, he threatens to return to a darker space.

"Stayed in trouble just to show these suckers I ain't lost it/ I'll probably do it again/ That's the funny thing about it."

However, he said "not at all" when asked if he'd do some of the things that caused him to face serious trouble before. "It's back there [in my mind]. It's a piece of my mind that I have to be honest with you and share with you. It's a thought. If that thought is not there, I'd never be able to check myself. I'd never be able to say, 'Be careful: You know what could happen.' If I totally discredited the idea, I could never check myself."

"My life, your entertainment," he chants on the hook. "My sorrow, your amusement/ Laugh at my pain and anguish/ So famous I could lose it."

"For a long time, people came home and got they TVs on, looked at the news, just to see how my situation developed and entertain themselves," he explained of the record. "In the meantime, it's my life. People on the computer saying, 'T.I. this, T.I. that. They just said T.I. couldn't do that.' They comment. They don't know nothing about T.I."

The message of "Collect Call" is clear: "Everybody loves you when you ballin'/ Don't nobody want you when you fallin'."

"That song is about when you're on top, everybody wants to ball with you, but when you're not, you probably wouldn't know who to call" explained Tip, who raps on the song, "Everybody talking about me like a n---a just died."

"I'm Illie" is an A-Town anthem that resonates with T.I.'s boasts about being the man in charge.

"Never been f---ed in the game, I'm celibate," he declares. The song's chorus goes, "N---as talk sh--, that's silly/ I run this city, clearly ... Tell that little n---a I'm busy: I'm illaaayy!"

"That's just cats saying they want that old Trap Muzik flow back," he said. "That nonstop, consistent punch-line-and-metaphor wordplay. I gave them that."

T.I. hasn't picked out a first single for the LP yet: He's still recording.

Dancing on Ice stars gear up for final

'Dancing on Ice' finalist Chris Fountain has played down his tag as favourite ahead of Sunday night's live final.
The 20-year-old 'Hollyoaks' star also insisted that his ice-hockey background does not give him an unfair advantage over the other contestants.
Fountain said: "I suppose it gave me a general awareness of the ice, but I still don't think I would have been scared to fall over because that's just the type of person I am - I throw myself into anything."
"I think I've tried some really difficult stuff and I don't feel like I've pushed myself less than anybody else. I don't feel like I've sat back on my experience and gone, 'I can do an easy routine and just sail my way through'."
Fellow finalist Suzanne Shaw said: "I think everyone's aware of how far they can push themselves."
"I know when to stop and I'm not stupid that I'll take it too far; that I will break something."
Former 'Coronation Street' star Zaraah Abrahams, who has also made the final of the show, said that she was skating for Michael Underwood, who was forced out of the show after he broke his ankle.
"They (Underwood and dance partner Melanie Lambert) often come down to our training and they get us going. They're so proud and we're so proud because we've done so well for them - we've got them into the final."

THE WAIFS

THE WAIFS

SunDirtWater (Compass): B-

Aboriginal Australian triad the Waifs have in full embraced the mainstream sounds of their adopted United States in this disappointing follow-up to 2002’s entrancing “Up Totally Night.” Recorded in Nashville with Medicine City’s pM school term players, there’s a dearth of the creative quirks that made their previous work so enjoyable. Excessively many of these folky pop songs sound like second-rate Sheryl Corvus, lukewarm Norah Jones or mediocre res publica blues. Still, Jolly Merce Cunningham and sisters Donna Mrs. Simpson and Vikki Irritant always deliver at least one killer caterpillar track: this time it’s the farfisa-organ punctuated “Quell,” the unity to download.
